A standard production package for expression in E. coli:

Amount< 20 mg, >95% purity, soluble protein <100 kDa single subunit, 2-4 weeks.

  • Phase I: Molecular Biology (if needed)

    • Codon Optimization: Redesigning the gene sequence to match preference for higher expression.

    • Gene Synthesis & Sub-cloning: Building the DNA and inserting it into an expression vector (e.g., pET system).

  • Phase II: Expression Testing (Pilot Study)

    • Strain Screening: Testing different strains (e.g., BL21).

    • Small-scale expression: Evaluating solubility and yield before full-scale production.

  • Phase III: Scale-up and Purification

    • Large-scale Culture: 1L to 8L of culture.

    • Purification: Usually two-step purification (e.g., 1. Affinity for tags, 2. Protein-specific polishing column).

  • Phase IV: Deliverables & QC

    • Purified Protein: In your specified buffer.

    • Quality Control Data: SDS-PAGE (purity), Western Blot (identity: anti-tag, specific: Ab tested and provided by client), and a Certificate of Analysis (CoA). 

  • Once the protocol is established, scale-up production can be implemented. $2000, 50-100 mg
